THYMOQUİNONE’NİN İNSAN MEME KANSERİ HÜCRE HATLARI ÜZERİNE ETKİLERİ

Abstract




Thymoquinone (TQ), çörek otunun aktif
maddesi olup, birçok kanser türlerinin tedavisinde kullanılmaktadır. Fakat
insan meme kanseri hücre hatlarında henüz bir çalışma bulunmamaktadır.
Dolayısıyla bu çalışmada; TQ’nın insan meme kanseri meme hatlarına etkisinin
olup olmadığının araştırılması amaçlanmıştır. Çalışmada; İnsan meme
adenocarcinoma hücreleri (MCF–7) Amerikan kültür koleksiyonundan (ATCC) elde
edilerek kullanıldı.  Thymoquinone n
serisel dilüsyonları hazırlanarak mililitrede 1x105 hücre olacak
şekilde 24 kuyucuklu kültür peleytlerinde 370C’de bir gece inkübe
edilerek, böbrek epitelyum hücreleri ile karşılaştırıldı. Sonuçlarımızda 250 µM
konsantrasyona kadar TQ’nın toksin etki göstermediği, MCF-7’nin 250 µM’da
üremesini engellediği, fakat bu konsantrasyonda böbrek epitelyum hücrelerine
etki etmediği gözlemlendi. Bulgularımız; TQ’nin hücre siklusunu durdurarak
kanser hücrelerini öldürdüğünü göstermektedir. 250 µM konsantrasyonda kanser
ücresi olmayan hücrelerde ise dayanıklılığı da görüldü.
